摘要 目的观察胶体金免疫电镜技术(CGIMT)在脑缺血—再灌注(IR)后血脑屏障(BBB)损伤大鼠脑组织超微结构及水通道蛋白-4(AQP4)表达观察中的应用效果。方法将18只SD雄性大鼠随机分为对照组、模型组和电针组各6只,模型组和电针组均采用改良大脑中动脉栓塞(MCAO)方法建立局灶性IR模型,且电针组于MCAO造模成功后5 min开始电针治疗;对照组不做特殊处理。采用CGIMT观察三组脑组织超微结构及AQP4表达变化。结果对照组和模型组脑组织超微结构基本正常、AQP4低表达;模型组胶质细胞萎缩,核蛋白和胞质蛋白成分松散、丢失,粗面内质网致密,AQP4表达增多。结论 CGIMT能较好的保存细胞的超微结构和抗原活性、准确显示AQP4表达部位,可用于IR后脑超微结构变化的研究。 Objective To observe the application effect of CGIMT in the observation of rat brain tissue ultrastructure of blood-brain barrier (BBB) injury after cerebral ischemia-reperfusion (IR) and aquaporin-4 (AQP4) expression. Meth- ods Eighteen SD male rats were randomly divided into 3 groups, 6 in each group: the control group, model group and electroacupuncture group (EA group). Focal cerebral IR models were successfully established by MCAO in the model and EA groups, and electroacupuncture therapy was used in the EA group after 5 minutes of MCAO model establishment; the control group without any treatment. The brain tissue ultrastructure and AQP4 expression were observed by using CGIMT. Results Brain issue uhrastructure in the control and EA group was basically normal, with low expression of AQP4. In the model group, colloid cells atrophied, the components of nucleus and plasmosin proteins were loose, granular endoplasmic reticulum compacted, and the expression of AQP4 were higher. Conclusions CGIMT could preferable preserve the ultra- structure of cells and activity of antigens, display the AQP4 expression location accurately. CGIMT can be applied into the study of brain tissue uhrastructure of BBB injury after cerebral IR.
