蕨麻乙醇提取物对大鼠急性心肌缺血再灌注损伤的保护作用及相关机制 被引量:5

Protective effects and mechanism of alcohol extract of Potentilla anserine L. on acute myocardial ischemia/reperfusion injury in rats

摘要 目的:研究蕨麻乙醇提取物对急性心肌缺血再灌注损伤的保护作用及机制。方法:雄性SD大鼠随机分为假手术对照组,模型组,蕨麻乙醇提取物干预组,阳性药物组。建立急性心肌缺血再灌注损伤模型,测定心功能变化,血清学检测乳酸脱氢酶(LDH)、肌酸激酶(CK)、超氧化物歧化酶(SOD)活性及丙二醛(MDA)、一氧化氮(NO)含量;透射电镜观察超微结构变化,免疫组化检测诱导型一氧化氮合酶(iNOS)表达。结果:与模型组相比,蕨麻乙醇提取物能够明显改善心功能指标及心肌细胞超微结构,降低LDH、CK水平,减少MDA、NO产生,提高SOD活性,抑制iNOS表达。结论:蕨麻乙醇提取物对急性心肌缺血再灌注所致心肌损伤具有保护作用,其可能是通过提高机体抗氧化能力,抑制NO生成而实现的。 bjective: To study the protective effects and mechanism of alcohol extract of Potentilla anserina L. on acute myocardial ischemia/reperfusion injury in rats. Methods: Male Sprague-Dawley rats were randomly divided into sham group and injury groups which were subdivided into model and diltiazem groups, intervention groups of alcohol extract of Potentilla anserina L. The index of cardiac functionhaemodynamics was recorded. Activity of LDH, CK, SOD and productions of MDA, NO in serum were measured. The ultra-construction change of mitochondrion and glycogen in myocardial cell was observed by electron microscope. The expression of iNOS was evaluated by immunohistochemistry. Results: Pretreatment with alcohol extract of Potentilla anserina L. could significantly improve homodynamic myocardial functional parameters after I/R injury, degrade the activities of LDH and CK, reduce myocardial ischemia-reperfusion injury caused by swelling of mitochondria vacuoles, and inhibit the production of MDA and NO, enhanced the activity of SOD, attenuated the expression of iNOS. Conclusion: Pretreatment with alcohol extract of Potentilla anserina L. could protect the myocardial function from acute myocardial ischemia and reperfusion injury in rats, which may be achieved by enhancing the activity of SOD, increasing the clearance of ROS, and attenuating the production of NO.
