摘要
目的:观察骨质疏松症大鼠骨骼、骨骼肌Ⅰ型胶原mRNA表达变化影响。方法:将120只雌雄各半的大鼠随机分为正常组、模型组、补肾组、健脾组、活血组和骨疏康组6个组。测定大鼠骨骼、骨骼肌Ⅰ型胶原mRNA表达。结果:①与正常组相比,模型组大鼠骨组织Ⅰ型胶原mRNA表达水平明显下降(P<0.05),与模型组相比,补肾组升高趋势最为显著(P<0.01),其次为健脾组(P<0.05)。②与正常组相比,模型组大鼠骨骼肌Ⅰ型胶原mRNA表达水平明显下降(P<0.01),与模型组相比,补肾组、健脾组升高趋势最为显著(P<0.01)。结论:①骨质疏松症的形成,可能与骨骼、骨骼肌Ⅰ型胶原表达的异常变化有关,印证了中医学骨痿"骨肉不相亲"的病机。②补肾、健脾方法通过调控骨质疏松症大鼠的骨骼、骨骼肌Ⅰ型胶原mRNA表达,对骨质疏松症具有一定的防治作用。
Objective: To investigate m RNA expression of Col l in the skeleton and skeletal muscle of osteoporosis rats. Methods: 120 rats (male/female=1:1) was randomly divided into 6 groups as following blank controlling group, model controlling group, invigorating the kidney group, invigorating the spleen group, promoting blood flow group and Gusukang group. We took the way of RT-PCR to examine the m RNA expression of Col I in the skeleton and skeletal muscle. Results: ①Compared in the result of mRNA expression of Col Ⅰ in the skeleton, model controlling group was significantly lower than blank controlling group (P〈0.05). Compared to the model controlling group, the invigorating the kidney group showed the greatest difference (P〈0.01) followed by invigorating the spleen group (P〈0.05). ②Compared in the result of mRNA expression of Col I in the skeletal muscle, model controlling group was significantly lower than blank controlling group (P〈0.01). Compared to the model controlling group, the invigorating the kidney group and invigorating the spleen group both showed the greatest difference (P〈0.01). Conclusion: ①The abnormal expression of Col I in the skeleton and skeletal muscle may plays an essential role in the procedure of osteoporosis forming. ②Treatment of invigorating the kidney and invigorating the spleen shows effective to cure osteoporosis probably by regulating the mRNA expression of Col I in the skeleton and skeletal muscle.
