壮筋续骨汤对大鼠骨折骨痂中骨形态发生蛋白-7和神经肽Y表达的影响 被引量:17

Effects of Zhuangjin Xugu Decoction on the expressions of BMP-7 and NPY of fracture callus in rats

摘要 目的:研究壮筋续骨汤对大鼠股骨骨折骨痂中骨形态发生蛋白-7(BMP-7)与神经肽Y(NPY)表达的影响,探讨其促进骨折愈合的作用。方法:成年健康雄性Wistar大鼠50只,应用线锯在股骨中段切断股骨制备骨折模型,不同剂量壮筋续骨汤水煎液灌胃治疗。X线观察骨折愈合情况,苏木精-伊红(HE)染色观察骨折的病理变化。免疫组织化学染色检测骨痂中BMP-7和NPY的表达。结果:经壮筋续骨汤治疗7-14d,X线片示大鼠股骨骨折线模糊,HE染色示骨折断端纤维肉芽组织逐渐转变为纤维性骨痂组织和软骨性骨痂。免疫组化显示,各剂量治疗组治疗7d后骨痂中成纤维细胞和成骨细胞BMP-7和NPY表达增强,第14天表达最高,与模型组比较差异有统计学意义(P<0.05)。结论:壮筋续骨汤可能通过提高骨痂组织成纤维细胞中BMP7和NPY的表达而促进骨折愈合。 Objective: To study the effects of Zhuangjin Xugu Decoction on the expressions of bone morphogenetic protein-7 (BMP-7) and neuropeptide Y (NPY) in callus of femoral fracture and the enhancing fracture healing in rats. Methods: Total of 50 adult healthy male Wistar rats were established fracture models by cutting femur transversely at middle point, and treated with Zhuangjin Xugu Decoction by gastric administration. The fracture healing condition was observed by X-ray and the pathological conditions of fracture healing was detected by HE staining. The expressions of BMP-7 and NPY in callus were detected by immunohistochemical assay. Results: After treatment 7-14 days, the femoral fracture line in X-ray film became unclear gradually. The fibrous-granulation tissue formed the fibrous and cartilaginous callus tissue successfully at the fracture- end. Immunohistochemical assay indicated that the expressions of BMP-7 and NPY in fibroblast osteoblast of callus tissues increased successfully at 7 days after treatment, peaked at 14 days, and more than those in femoral fracture model group (P〈0.05). Conclusion: Zhuangjin Xugu Decoction could enhance the fracture healing by up-regulating the BMP-7 and NPY expressions in fibroblast of callus tissues in rats.
