摘要
目的:观察益肺清化颗粒对Lewis肺癌小鼠血清中的血管生成因子:血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、血管抑素(AS)、内皮抑素(ES)的影响,以期探讨益肺清化颗粒对肿瘤新生血管的作用机制及抑瘤机制。方法:通过建立小鼠Lewis肺癌模型,并给与不同药物干预,造模成功后,留取小鼠眼球血,用酶联免疫吸附试验(ELISA)方法检测以上4种血管生成因子的含量。结果:益肺清化颗粒低、中剂量组、高剂量组、吉非替尼组、吉非替尼+益肺清化颗粒中剂量组、环磷酰胺(CTX)组VEGF的表达均较模型组显著降低(P<0.05,P<0.01);吉非替尼组、吉非替尼+益肺清化颗粒中剂量组、CTX组对AS的影响与模型组比较差异有统计学意义(P<0.05,P<0.01);益肺清化颗粒中、高剂量组、吉非替尼+益肺清化颗粒中剂量组、吉非替尼组、CTX组对ES的影响与模型组比较差异有统计学意义(P<0.05,P<0.01)。结论:益肺清化颗粒能降低血管生成促进因子VEGF的表达及提高ES的表达,益肺清化颗粒治疗肺癌的可能作用机制为:降低血管生成促进因子VEGF的表达,升高ES的表达,进而发挥抑瘤作用。
Objective: Observation of the effect of Traditional Chinese Medicine Yifei Qinghua Granular on vascular endothelial growth factor in Lewis Lung Cancer mouse serum including vascular endothelial cell growth factor, basic fibroblast growth factor, Angiostatin and Etldostatin, to explore the action mechanism and antitumo mechanism of Yifei Qinghua granular on tumor microvasculature. Methods: By establishing the model of Lewis Lung Cancer, and give them different drugs intervention. After establishment of the successful model, taking the blood of mice eyeball to detect the content of the above four angiogenic factors by ELISA method. Results: Compared with the model group, the expression of VEGF decreased significantly in Yifei Qinghua granular low dose group and middle dose group, Yifei Qinghua granular high dose group, Gefitinib group, Gefitinib+Yifei Qinghua granular middle dose group, and CTX group (P〈0.05, P〈0.01); Compared with the model group, the expression of Angiostatin in Gefitinib group, Gefitinib+Yifei Qinghua granular middle dose group, CTX group were statistically significant (P〈0.05, P〈0.01). The expression of Endostafin in Yifei Qinghua granular middle and hige dose group, Gefitinib+Yifei Qinghua granular middle dose group, Gefitinib group and CTX group were statistically significant (P〈0.05, P〈0.01). Conclusion: Yifei Qinghua granular could reduce the expression of vascular endothelial growth factor VEGF which was a angiogenesis promoting factor, and increase the expression of Endostatin which was a angiogenesis inhibiting factor. The action mechanism of Yifei Qinghua granular in lung cancer may reduce the expression of Angiogenesis promoting factor VEGF and increase the expression of Angiogenesis inhibitor Endostatin.
