摘要
目的研究蔓性千斤拔抗氧化活性部位及其化学成分。方法采用体外1,1-二苯基-2-苦基肼(DPPH)抗氧化模型对蔓性千斤拔提取物及其不同溶剂萃取部位的抗自由基活性进行评价;采用硅胶、Sephadex LH-20柱色谱及半制备HPLC等方法对活性部位的化学成分进行分离,根据理化常数和波谱数据鉴定化合物结构。结果氯仿萃取部位抗氧化活性最强,正丁醇萃取部位次之。从活性部位中共分离得到21个化合物,分别鉴定为:棕榈酸(1),羽扇豆醇(2),β-谷甾醇(3),咖啡酸二十四烷酯(4),对甲氧基苯丙酸(5),木蜡酸(6),5,7-二羟基-6,8-二异戊烯基色原酮(7),flemiphilippinin E(8),5,7,4′-三羟基-3′-甲氧基-6,8-二异戊烯基异黄酮(9),6,8-二异戊烯基柚皮素(10),5,7,4′-三羟基-6,8-二异戊烯基异黄酮(11),pomiferin(12),5,7,3′,4′-四羟基-6,8-二异戊烯基异黄酮(13),desmoxyphyllin A(14),5,7,2′,4′-四羟基-8-(1,1-二甲基-2-丙烯基)异黄酮(15),5,7,4′-三羟基-2′-甲氧基异黄酮(16),ψ-赝靛素(17),芒柄花素(18),槲皮素(19),染料木素(20)和染料木苷(21)。结论化合物1,4,6,14-17为首次从该属植物中分离得到。
OBJECTIVE To investigate the chemical constituents of antioxidative fractions in roots of Moghania philippinensis ( Merr. et Rolfe) Li.. METHODS The antioxidant activities of the extract and fractions were evaluated by determining their ability to scavenge DPPH radicals. The compounds were isolated by chromatographic techniques with silica gel, Sephadex LH -20 and semi-preparative HPLC. Their structures were identified by analysis of physical and spectral evidence, and confirmed by comparison of their spectral data with reported values in the literature or those of authentic samples. RESULTS The chloroform and n-butanol extracts showed strong scavenging activity against 2, 2-diphenyl-1-picrylhydrazyl(DPPH). Twenty-one compounds were isolated and identified as palmitic acid ( 1 ), lupeol ( 2 ), β-sitosterol ( 3 ), 3′, 4 ′-dihydroxy-trans-cinamic acid tetracosyl ester ( 4 ), 3- ( 4-methoxyphenyl ) propionic acid ( 5 ), n-tetracosanoic acid (6), 5, 7-dihydroxy-6, 8-diprenylchromone (7), flemiphilippinin E (8), 5, 7, 4′-trihydroxy-3′-methoxy-6, 8-diprenylisoflavone (9), 6, 8-dipreny naringenin ( 10), 5, 7, 4′-trihydroxy-6, 8-diprenylisoflavone ( 11 ), pomiferin ( 12 ), 5, 7, 3′, 4′-tetrahydroxy-6, 8-diprenylisoflavone (13), desmoxyphyllin A (14), 5, 7, 2′, 4′-tetrahydroxy-8-(1, 1-dimethylprop-2-enyl) isoflavone ( 15 ), 5, 7, 4′-trihydroxy-2′-methoxy-isoflavone ( 16 ), ψ-baptigenin ( 17 ), formononetin ( 18 ), quercetin ( 19 ), genistein (20) and genistin(21), respectively. CONCLUSION Compounds 1,4, 6, 14 -17 are found from this genus for the first time.
