摘要
本研究通过减少柞蚕蛹血淋巴含量而添加不同配比灵芝多糖、壳寡糖溶液优化松毛虫赤眼蜂体外培育的人工卵液配方。当血淋巴含量从40%降低到30%而添加比例分别为10:0,4:1,3:2,1:1,2:3,1:4和0:10的灵芝多糖与壳寡糖溶液时,各配方与对照比较,均能显著促进松毛虫赤眼蜂的生长发育;除含有相同灵—壳配比(4:1)的人工卵液无明显差别外,含30%血淋巴的新配方培育的松毛虫赤眼蜂各生物学指标和质量参数均明显优于含25%血淋巴的新配方。分别筛选出A4(30%柞蚕蛹血淋巴,30%鸡蛋黄,20%鲜牛奶,10%N’s盐,5%灵芝多糖溶液,5%壳寡糖溶液)和B2(25%柞蚕蛹血淋巴,30%鸡蛋黄,20%鲜牛奶,10%N’s盐,12%灵芝多糖溶液,3%壳寡糖溶液)两个能显著改善基础人工卵液的新配方,其中A4更适于大量体外培育松毛虫赤眼蜂。
Artificial medium for rearing Trichogramma dendrolimi Matsumura in vitro was optimized by reducing the content of pupal hemolymph ofAntheraeapernyi (Guerin-Meneville), and adding different ratio of Ganoderma lucidum polysaceharides and chitosan oligosaccharide solution. Compared to CK, when hemolymph was reduced from 40% to 30% and Ganoderma lucidum polysaccharides and chitosan oligosaccharide solution was increased by ratio of 10:0, 4: 1, 3:2, 1 : 1, 2:3, 1:4 and 0:10, these new artificial media significantly improved developmental parameters. T. dendrolimi female wasps did not laid eggs in media supplemented with Ganoderma lucidum polysaccharides and chitosan oligosaccharide solution at ratio of 3 : 2, 2: 3 and 1 : 4, but there were no significant differences in developmental parameters of the wasp, including rates of parasitism, pupation, adult emergence, and normal adults compared to the media with Ganoderma lucidum polysaccharides and chitosan oligosaccharide solution at ratio of 15:0, 4: 1. T. dendrolimi developed well on the new media with 30% hemolymph than on the media with 25% hemolymph, except on medium with Ganoderma lucidum polysaccharides and chitosan oligosaccharide solution at ratio of 4: 1. The results showed that insect hemolymph was a key component of the artificial media. Two artificial media A4 (30% hemolymph, 30% egg yolk, 20% fresh milk, 10% N's salt solution, 5% Ganoderma lucidum polysaccharides solution, 5% chitosan oligosaccharide solution) and B2 (25% hemolymph, 30% egg yolk, 20% fresh milk, 10% N's salt solution, 12% Ganoderma lucidum polysaccharides solution, 3% chitosan oligosaccharide solution) were screened out, and media A4 had the potential to be used in mass production of T. dendrolimi.
