瑞芬太尼预处理对糖尿病大鼠心肌缺血-再灌注时心肌细胞凋亡的影响 被引量:3

Effects of remifentanil preconditioning on cardiomyocyte apoptosis induced by myocardial ischemia-reperfusion in diabetic rats

摘要 目的探讨瑞芬太尼预处理对糖尿病大鼠心肌缺血-再灌注损伤的影响。方法将成功建立的糖尿病大鼠模型36只随机均分为三组:假手术组(Sham组)、缺血-再灌注组(IR组)和瑞芬太尼预处理组(RE组),每组12只。再灌注120min时,计算心肌梗死面积及采用Western blot法测定心肌组织Bax和Bcl-2蛋白的表达。结果与Sham组比较,IR组Bax蛋白的表达明显升高,Bcl-2蛋白的表达明显降低(P<0.05);与IR组比较,RE组心肌梗死面积和心肌组织Bax蛋白的表达均明显降低(P<0.05),而Bcl-2的表达明显增加(P<0.05)。结论瑞芬太尼预处理可能通过调节Bax和Bcl-2蛋白的表达,减少细胞凋亡,从而减轻糖尿病大鼠的心肌缺血-再灌注损伤。 Objective To investigate the effects of remifentanil preconditioning on myocardial ischemic reperfusion injury in diabetic rats. Methods Diabetic rats were randomly divided into there groups (n= 12) : Sham group: received sham operation in which the coronary suture was passed, but not tied; group IR received isehemia for 30 min followed by 120 min reperfusion; group RE received injection of remifentnail before ischemia, remifentnail was injected intravenously (6/,g. kg-~ ~ rain-1) during 5 min for three times and interspersed with 5 min drugs-free periods. The rats were sacrificed for infarct size assessment to determine of Bax and Bcl-2 expression by Western blot. Results Compared wih group Sham, group IR significantly increased the Bax expression, and reduced Bcl-2 expression; group RE significantly reduced the infarct area and Bax expression, and increased the expression of Bel-2 compared with group IR. Conclusion Remifentanil preconditioning could reduce myocardial ischemic reperfusion injury in diabetic rats by regulated Bax, Bcl-2 expression and inhibitting cardiomyocyte apoptosis.
