摘要
目的评价不同浓度CD46预先给药对脂多糖(lipopolysaccharide,LPS)诱导的鼠原代小胶质细胞补体蛋白表达的影响。方法培养原代小胶质细胞,取分离纯化的小胶质细胞接种于24孔板,随机分为4组(n=6):正常对照组、LPS组、低浓度CD46组和高浓度CD46组。采用噻唑蓝法检测细胞活性,采用酶联免疫吸附(ELISA)法测定培养上清液补体C3浓度。结果 LPS组、低浓度CD46组、高浓度CD46组补体C3表达均较正常对照组增加(P<0.01);低浓度CD46组、高浓度CD46组补体C3表达均较LPS组减少,且高浓度CD46组补体C3表达较低浓度CD46组减少(P<0.05)。结论 CD46可明显抑制LPS诱导的小胶质细胞激活,显著减少补体C3释放。
Objective To evaluate the effect of different concentrations of CD46 pretreatment on protein C expression in primary culture of microglia cells induced by Lipopolysaccharide(LPS) in rats.Methods The primary microglia cells were cultured.Separated and purified microglia cells were vaccinated in 24 orifices plate,and were randomly divided into 4 groups(n = 6) : control group,LPS group,low concentration of CD46 group and high concentration of CD46 group.Cytoactive was detected by methylthianolyldipheyl-tetrazolum bromide(MTT),and the concentration of supernate fluid complement C3 was measured by enzyme linked immunosorbent assay(ELISA).Results Expressions of complement C3 in LPS group,low concentration of CD46 group and high concentration of CD46 group were increased than that in control group(P 0.01);expressions of complement C3 in low concentration of CD46 group and high concentration of CD46 group were reduced than that in LPS group,and expression of complement C3 in high concentration CD46 group was reduced than that in low concentration CD46 group(P 0.05).Conclusion CD46 may obviously restrain microglia cell-stimulating induced by LPS,and reduce release of complement C3.
