摘要
以嫩芽、茎尖为外植体,研究巨尾桉DH3229的组织培养及育苗技术。实验证明,不同的激素组合对巨尾桉DH3229外植体的诱导、生根有不同的影响。最适合诱导的增殖培养基为MS+6-BA 0.5mg/L+NAA 0.2mg/L,其诱导出来的植株生长最快速,分枝最多,植株最高;最适合的生根培养基为MS+NAA 1mg/L+6-BA 0.2mg/L,生根率达94%,主根粗壮,平均长3根根须。但是移植到基质为黄土的培养杯中育苗时,成活率最高的培养基是MS+NAA0.5mg/L+6-BA 0.2mg/L,成活率达93%。
The tender buds were used as explants to study the tissue culture and genetic improvement of Eucalyp- tus grandisXEucalyptus urophylla DH3229 in the experiment.The results were as follws: The optimal subculture medium was MS+6-BA 0.5mg/L+NAA 0.2mg/L, in which the plants grew expeditiously and strongly.The optimal rooting medium for test-tube plants was MS+NAA lmg/L+6-BA 0.2mg/L,in which the rooting rate reached 94% and per plant could produce 3 roots.But the best optimum medium for plant transplantation was MS+NAA 0.5mg/ L+6-BA 0.2mg/L,in which the transplanting survival rate of the plants reached 93%.
