糖网明含药血清对高糖条件下视网膜神经节细胞NO、ROS水平的影响

EFFECTS OF TANGWANGMING ON NO AND ROS LEVELS OF RETINAL GANGLION CELLS CULTURED IN HIGH GLUCOSE

摘要 目的:观察益气养阴活血中药——糖网明对高糖条件下大鼠视网膜神经节细胞一氧化氮(NO)及活性氧簇(ROS)的影响。方法:体外培养新生大鼠视网膜神经节细胞(RGCs)。实验分为正常对照组、正常中药干预组、高糖组及高糖中药干预组。以电子自旋共振法(ESR)检测各组一氧化氮及活性氧的释放来反映氧化应激程度。结果:高糖作用导致RGCs释放NO、ROS增多,糖网明干预能显著地降低NO、ROS水平(P<0.01)。结论:益气养阴活血中药糖网明能在一定程度上减轻高糖对视网膜神经节细胞的氧化应激损伤,从而发挥其对RGCs的保护作用。 Objective:To observe the effects of tangwangming on NO(nitrogen monoxide) and ROS(reactive oxygen species) levels of rats’retinal ganglion cells cultured in high glucose.Methods:Experiment was divided into four groups,blank control group,tangwangming group,high glucose group and tangwangming plus high glucose groups,rats’retinal ganglion cells of each group were cultured.NO and ROS which showed oxidative stress in retinal ganglion cells were determined by ESR(electron spin resonances) .Results:The release of NO and ROS in retinal ganglion cells which cultured in high glucose was increased obviously,that was reduced significantly after intervening with tangwangming(P < 0.01) .Conclusion:Tonifying qi and nourishing yin Chinese herbs decoction tangwangming can protect retinal ganglion cells by reducing oxidative stress injury caused by high glucose.
