Functional defect of regulatory T cells in peripheral blood of patients with multiple sclerosis

摘要 目的:探讨多发性硬化患者外周血调节性T细胞(Treg)的变化及其转录因子Foxp3的表达。方法:收集58例多发性硬化(MS)患者及24例正常人外周血,通过流式细胞术检测CD4+CD25+T细胞,RT-PCR检测Foxp3的表达。结果:MS患者外周血中Treg细胞数量较正常人无差异(P>0.05),而Foxp3表达明显减低(P<0.05)。结论:MS患者外周血中Treg细胞数量较正常人无明显变化,但其功能明显受损,是导致自身免疫性疾病发生的原因之一。 Objective:To detect regulatory T cells and Foxp3 expression in peripheral blood of patients with multiple sclerosis(MS).Methods: A total of 58 MS patients and 24 healthy people were included in this study,the number of CD4+CD25+ regulatory T cells in peripheral blood were detected by flow cytometry,Foxp3 expression were analyzed by real-time fluorescent quantitative RT-PCR.Results: There was no difference of the number of Treg cell between MS patients and healthy people,but the expression of Foxp3 were significantly decreased in MS patients(P0.01).Conclusion: The function of Treg cells were severely impaired in MS patients although its number had not changed compared with healthy people,which could contribute to the onset of this autoimmune disease.
