摘要
目的:探讨雄黄对NB4细胞(维甲酸敏感的急性早幼粒细胞白血病细胞株)组织因子(tissue factor,TF)和组织型纤溶酶原激活物(tissue-plasminogen activator,t-PA)表达的影响及机制。方法:用300μg/L雄黄与NB4细胞共同孵育6、12、24、48、72h。ELISA法、RT-PCR检测未处理及雄黄处理后NB4细胞TF、t-PA蛋白表达及mRNA转录的变化,检测未处理的MR2、HL-60、K562细胞的TF、t-PA蛋白表达作为对照。RT-PCR检测未处理及雄黄处理后NB4细胞PML/RARαmRNA转录的变化。结果:300μg/L雄黄可呈时间依赖方式下调NB4细胞TF蛋白表达及mRNA的转录,但不能影响t-PA蛋白表达及mRNA的转录。雄黄不影响NB4细胞PML/RARαmRNA的转录。结论:下调NB4细胞TF的表达从而降低NB4细胞促凝活性可能是雄黄改善急性早幼粒细胞白血病(APL)患者弥散性血管内凝血(DIC)早期出血症状的主要机制之一。但雄黄对NB4细胞TF表达的调节作用可能与融合基因无关。雄黄对t-PA表达的影响有待进一步研究。
Objective:To investigate the effect of realgar on tissue factor(TF) and tissue-plasminogen activator(t-PA) expression and mRNA transcription in acute promyelocytic leukemia(APL) cell line-NB4 cells.Methods:NB4 cells were treated with 300μg/L realgar.TF and t-PA antigen was detected by ELISA and mRNA by RT PCR.PML/RARα mRNA was detected by RT-PCR.Results:Realgar could down-regulate the expression of TF in a time dependent manner.The expression of t-PA was not influenced by realgar in our research.The transcription of PML/RARα mRNA was not affected by realgar.Conclusion:Down-regulating the TF of NB4 cells by realgar may be one of the mechanisms of its improvement on DIC-related hemorrhage of APL patients.The effect of realgar on t PA expression needs further research.
