过量扑热息痛减少小鼠肝中GSH的机制探讨 被引量:2

摘要 目的探讨过量扑热息痛(AAP)减少急性肝损伤小鼠肝组织中还原型谷胱甘肽(GSH)含量的可能新的机制。方法雄性昆明种小鼠一次性腹腔注射AAP 350 mg/kg,18 h后取血测定血清中丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)的含量,取肝组织测定总谷胱甘肽和氧化型谷胱甘肽(GSSG)含量,并计算GSH含量和GSH/GSSG比值,以及谷胱甘肽过氧化物酶,谷胱甘肽-S-转移酶和谷胱甘肽还原酶(GR)的活性,同时取肝组织进行病理形态学检查。结果小鼠给予过量AAP后能明显升高血清ALT和AST含量(P<0.01),光学显微镜下可见肝组织中有不同程度的炎细胞浸润和肝细胞变性及坏死,肝组织中的GSH含量、GSH/GSSG比值和GR活性均明显降低(P<0.05或P<0.01)。结论过量AAP引发的GR活性下降可导致GSH的进一步减少,这可能是过量AAP引起肝损伤的新的机制。
