摘要
目的探讨白细胞介素6(IL-6)及其受体在胰腺癌细胞的表达及IL-6对胰腺癌细胞产生血管内皮生长因子C(VEGF-C)的调节。方法采用Northern杂交法(Northern blot)测定胰腺癌细胞Capan-1、Colo-357和5H中IL-6及其受体和VEGF-C mRNA的表达;以同法检测胰腺癌细胞Capan-1和Colo-357受IL-6刺激前后VEGF-CmRNA的表达及变化。结果 IL-6及其受体和VEGF-C mRNA在胰腺癌细胞Capan-1、Colo-357和5H中均有不同程度的表达。IL-6对胰腺癌细胞Capan-1表达VEGF-C mRNA的调节:以50μg/L浓度的IL-6刺激Capan-1细胞3小时、6小时后,与对照组(0小时组)相比,2.4kb片段的VEGF-C mRNA的条带强度减弱17%~40%,IL-6对胰腺癌细胞Capan-1VEGF-C mRNA的表达有抑制作用(P<0.05),并随作用时间呈正相关;IL-6对胰腺癌细胞Colo-357表达VEGF-C mRNA的调节:以50μg/L浓度的IL-6刺激Colo-357细胞3小时、6小时后,与对照组(0小时组)相比,2.4kb片段的VEGF-C mRNA的条带强度增强1.2~1.6倍,IL-6对胰腺癌细胞Colo-357VEGF-C mRNA的表达有促进作用(P<0.05),但与作用时间无明显关系。结论 IL-6及其受体和VEGF-C mRNA在胰腺癌细胞Capan-1、Colo-357和5H中有不同程度的表达;IL-6抑制胰腺癌细胞Capan-1VEGF-C mRNA的表达,从而可能抑制胰腺癌淋巴结转移;IL-6促进胰腺癌细胞Colo-357VEGF-C mRNA的表达,从而可能促进胰腺癌淋巴结转移。
Objective To explore the expression of cytokine interleukin-6(IL-6) and its receptor in pancreatic cancer cells,and to study the regulation role of IL-6 in the expression of vascular endothelial growth factor C(VEGF-C) in cultured human pancreatic cancer cell lines.Methods We used Northern blot to measure the expression of IL-6,IL-6 receptor and VEGF-C mRNA in pancreatic cancer cells(Capan-1,Colo-357 and 5H),and used the same method to detect IL-6 regulation to VEGF-C mRNA expression in cultured pancreatic cancer cells Capan-1 and Colo-357.Results Northern blot analysis revealed the presence of the 1 128 bp IL-6 mRNA transcript,3 319 bp IL-6 receptor mRNA transcript and 2.4 kb VEGF-C mRNA transcript in all 3 tested cell lines.IL-6 regulated VEGF-C mRNA expression in pancreatic cancer cell Capan-1:the levels of VEGF-C gene were 17%-40% decrease in Capan-1 cell line by stimulation with IL-6(50 μg/L) for 3 h,6 h(P0.05),and the expression was increased with treatment time.IL-6 regulated VEGF-C mRNA expression in pancreatic cancer cell Colo-357:the levels of VEGF-C gene were 1.6-1.2 fold increase in Colo-357 cell line by stimulation with IL-6(50 μg/L) for 3 h,6 h(P0.05),however,the expression was no connection with the treatment time.Conclusion Northern blot analysis revealed the presence of IL-6,its receptor and VEGF-C mRNA in pancreatic cancer cells Capan-1,Colo-357 and 5H.IL-6 inhibited the expression of VEGF-C mRNA in pancreatic cancer cell Capan-1 and it suggests that IL-6 may inhibit lymph node metastasis of pancreatic cancer;IL-6 increased the expression of VEGF-C mRNA in pancreatic cancer cell Colo-357 and it suggests that IL-6 may facilitate lymph node metastasis of pancreatic cancer.
