摘要
目的:研究酸枣仁总皂苷对缺血再灌注损伤大鼠心律失常和心肌组织的保护作用及其对氧自由基的影响。方法:将24只Wistar雄性大鼠随机分为假手术组、模型组和酸枣仁总皂苷组(20mg.kg-1.d-1),采用结扎(15min)和松开(60min)左冠状动脉前降支的方法制备用心肌缺血再灌注损伤模型。记录再灌注60min期间II导联心电图,并对心律失常进行评分;常规HE染色观察心肌组织的病理改变;检测血清中乳酸脱氢酶(LDH)活性,心肌组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性。结果:模型组中心肌组织病理损伤及心律失常发生明显,LDH活性及MDA含量显著升高(P<0.01),SOD活性显著下降(P<0.01)。与模型组比较,酸枣仁总皂苷干预后,心肌组织病理改变减轻,心律失常评分显著降低(P<0.01),LDH活性及MDA含量显著下降(P<0.01),SOD活性显著提高(P<0.01)。结论:酸枣仁总皂苷A对缺血再灌注损伤大鼠心律失常具有保护作用,其机制可能与其抗氧自由基有关。
Objective: To study the effect of semen ziziphi spiosaeand on arrhythmia and oxygen free radical of myocardial ischemia reperfusion injury in rat.Methods: 24 Wistar male rats were randomly divided to three groups: sham operation group,model group and semen ziziphi spiosaeand group(in the dose of 20mg · kg-1· d-1). The animal model of myocardial isehemia reperfusion injury were established by ligating the left coronary arteries for 15min and then loosing for 60min.Arrhythmia was scored according to II-lead electrocardiograme.HE staining was used to observe the pathological changes of myoeatdium.Lactate dehydrogenase in serum,the malondialdehyde and superoxide dismutase in myocatdium tissue were measured after reperfusion. Resuhs:Ischemic injury changes of the myocatdium and arrhythmia occurred obviously (P〈 0.01 ), the activity of LDH in serum and the content of MDA in myocardium increased significantly(P〈0.01 ), the activity of SOD in myocardium decreased significantly Compared with sham operation group(P〈0.01 ). Compared with model group, semen ziziphi spiosaeand relieved the pathology changes in myocatdium,decreased arrhythmia score(P〈0.01 ), the activity of LDH in serum and the content of MDA in myocardium significantly(P〈0.01 ),increased the activity of SOD in myocardium obviously(P〈0.01 ). Conclusion: Semen ziziphi spiosaeand has a protective effect antiarrhythmia of myocardial ischemia reperfusion injury in rats,the mechanism may be related with antioxygen free radical.
