细胞因子对白血病抑制因子在小鼠孕早期子宫内膜表达的调节 被引量:17

The modulation of cytokines on the expression of leukemia inhibitory factor in the endometrium of early pregnant mice

摘要 本研究观察了白细胞介素 - 1(IL - 1)、白细胞介素 - 6 (IL - 6 )、单、粒 -细胞集落刺激因子 (GM- CSF)对子宫内膜白血病抑制因子 (LIF)表达的调节 ,探讨围着床期 LIF表达的调节机制。给妊娠 2~ 4 d的小鼠分别腹腔注射不同剂量的 IL - 1、白介素 - 1受体拮抗剂 (IL- 1ra)、IL- 6、GM- CSF,对照组注射 PBS,用免疫印迹法逆转录 -聚合酶链反应进行子宫内膜 LIF蛋白和 m RNA的半定量分析。结果 :对照组 LIF蛋白水平为 39.6 4± 9.95,与对照组比 ,IL - 14μg/kg剂量组明显升高 (P<0 .0 1) ;IL- 6和 GM- CSF 0 .8和 4μg/kg剂量组均明显升高 (分别为 P<0 .0 5,P<0 .0 1)。对照组 L IFm RNA水平为 1.10± 0 .2 1、IL-1、IL- 6和 GM- CSF4 μg/kg剂量组明显高于对照组 (分别为 P<0 .0 5,P<0 .0 5,P<0 .0 1) ,IL - 1ra 2 0μg/kg剂量组与对照组相比明显下降 (P<0 .0 5)。表明 IL- 1、IL - 6、GM-CSF对孕早期小鼠子宫内膜 LIF转录和转录后水平均有上调作用 ,可能是 L Leukemia inhibitory factor (LIF)is an essential factor for implantation. Little in known about the regulating mechanism of LIF expression in the endometrium. The aim of the present study is to observe the effects of interleukin(IL) 1,IL 6 and granulocyto macrophage colony stimulating factor(GM CSF)on the expression of LIF in the endometrium of early pregnant mice. Materials and methods:Pregnant mice were randomly allocated to 9 groups Experimental groups were daily injected with IL 1, IL 1ra, IL 6,GM CSF in different concentrations and PBS for control from day 2 to 4 of pregnancy. The mice were killed 12 h after the last injection, the endometrium was collected for the measurenment ofLIF protein and mRNA by immunoblot method and reverse transcription polymerase chain reaction. Results: Protein levels of LIF were signficantly higher in groups of IL 1 4 μg/kg (95.77±10.09, P <0 01) ,IL 6 0.8 μg/kg(68.47±10.80, P <0.05),IL 6 4 μg/kg( 81.54 ±10.79, P <0.01),GM CSF 0.8 μg/kg(86.66±16.14, P <0.05),GM CSF 4 μg /kg(124.17±20.87, P <0.01)than in the control group (39.64±9.95).The levels of LIFmRNA in the endometrium were significantly higher in the groups of 4 μg/kg IL 1(1.96±0.45, P <0.05),IL 6(2.38±0.54, P <0.05)and GM CSF(3.24±0.54, P < 0.01) ,and decreased significantly in the group of 20 μg/kg IL 1ra(0 62±0 09, P <0 05)than that in the group of control(1.01±0.21). Conclusion: Under stimulating by IL 1,IL 6 and GM CSF,the expression of LIF in the endometrium of early pregnant mice was increased at the levels of transcription and post transcription. They may be the potential up regulating factors in the expression of LIF in the peri implantation endometrium.
