摘要
目的 探讨T细胞免疫核糖核酸 (iRNA)是否可以通过调节机体免疫功能而影响关节炎病理生理过程 ,为类风湿关节炎治疗探索一条新路。方法 利用大鼠佐剂性关节炎 (AA)动物模型 ,腹腔注射T细胞免疫核糖核酸 ,观察关节体积、关节指数及关节病理变化。结果 注射 10d后 ,关节体积、关节指数与关节炎对照组比较差异均有显著性 (P <0 0 5 ) ,病理结果显示滑膜组织增生及炎症细胞浸润均较关节炎对照组明显减轻。结论 T细胞iRNA调节免疫系统功能抑制关节炎病理生理过程 ,其机制可能是iRNA通过诱生大量细胞因子的作用。
Objective To study the effect of T lymphocyte immunized RNA (iRNA) in adjuvant arthritis (AA) in rats.Methods Arthritis was produced in Wistar rats after single injection of Freund′s complete adjuvant (FCA).After 14 days injection,the rats which had arthritis were given intraperitoneal injection of iRNA.Then the severity of arthritis was inspected by surveying the volume of paw and the arthritic index.Results The group treated with iRNA developed milder arthritis than the arthritic rats without it 10 days later (P<0 05),and the difference was more significant 15 days later (P<0 01).The arthritic index of the rat treated with iRNA was (8 6±1 9),and that of the other arthritic group was (10 6±2 1).The difference between the two groups was significant (P<0 05).Histopathology showed that the less synovial hyperplasia and the fewer infiltration of inflammatory cells in the group treated with iRNA than without it.Conclusion Administration of iRNA can inhibit the development of AA.
