摘要
目的:探讨TMPRSS4基因对结肠癌细胞增殖及迁移的影响。方法 :采用免疫组化方法检测结肠直肠癌组织和正常肠上皮中TMPRSS4的表达,并应用RT-PCR和Western印迹法检测结肠癌细胞株中TMPRSS4的表达,通过RNAi方法下调结肠癌细胞SW480中TMPRSS4的表达后,进一步采用CCK-8法、细胞划痕和transwell迁移试验检测细胞增殖、运动及迁移能力的改变。通过流式细胞仪检测TMPRSS4下调后对细胞凋亡的影响。结果:结肠直肠癌高表达TMPRSS4;肠癌细胞株SW480在mRNA及蛋白水平均高表达TMPRSS4。下调TMPRSS4能抑制SW480的增殖能力(P<0.05),并诱导细胞凋亡增加(P<0.05)。此外,下调TMPRSS4降低SW480细胞的迁移能力(P<0.05)。结论:在高表达细胞株中下调TMPRSS4有助于抑制肠癌细胞增殖,增加细胞凋亡并降低细胞的迁移能力。
Objective To investigate the effect of TMPRSS4 on the cell proliferation and migration of SW480 colorectal cancer cells. Methods The expression of TMPRSS4 in colorectal cancer and normal mucosa was investigated by immunohistochemistry. The expression of TMPRSS4 in colon cell lines was detected by RT-PCR and Western blot. RNAi was used to down-regulate the TMPRSS4 expression in colon cancer cell SW480. Cell proliferation, motility and migration were determined by cell counting CCK-8 assay, wound healing assay and transwell migration assay, respectively. Cell apoptosis after TMPRSS4 down-regulation was measured by flow cytometry. Results TMPRSS4 was highly expressed in colorectal cancer tissues. Colon cancer cell line SW480 highly expresses TMPRSS4 both at mRNA and protein level. Down-regulation of TMPRSS4 expression inhibited the cell proliferation and induced cell apoptosis of colon cancer SW480 cells (P〈0.05). Moreover, TMPRSS4 down-regulation decreased migration in SW480 cells (P〈0.05). Conclusions Down-regulation of TMPRSS4 can inhibit colorectal cell growth, increase cell apoptosis and decrease cell migration.
