转化生长因子β调节培养的人角膜基质细胞MMP-2和MMP-9的分泌 被引量:4

摘要 目的观察转化生长因子6(TGF-β)对培养的人角膜基质细胞分泌MMP-2和MMP-9的调节作用,探讨TGF-β治疗非感染性角膜溃疡的潜在可能性。方法采用酶谱法检测人角膜基质细胞MMP-2和MMP-9的分泌,并观察 TGT-β对 MMP-2和 MMP-9分泌的调节作用。结果培养的人角膜基质细胞分泌 MMP-2和 MMP-9,TGF-β抑制MMP-9的活性,且随TGF-β质量浓度增加而增强,随TGF-β作用时间延长而增强;对MMP-2的活性,TGF-β有较弱的促进作用。结论TGF-β作为一种MMP-9的抑制剂,在促进非感染性角膜溃疡愈合方面很有潜力。 Objective To observe modulation of TGF-β for secretion of MMP-2 and MMP-9 by cultured human corneal keratocytes and investigate the possibility of treatment of non-infectious corneal ulcer with TGF-β. Methods The second~ third passage cells were seeded at proper density, incubated with TGF-βof different concentration (0, 0. 1 1, 5 ng/ml) or for different time (24, 48, 72h). The supernatants were collected and checked. Zymography was applied to test secretion of MMP-2 and MMP-9 by human corneal keratocytes. To observe the TGF-β's effect on the secretion, the zymography results were examined by densitometric analysis. Results Cultured human corneal keratocytes secreted MMP-2 and MMP-9. TGF-β inhibited the activation of MMP-9 and the inhibition was dose-dependent and time-dependent. TGF-β mildly promoted the activation of MMP-2. Conclusion Cultured human corneal keratocytes secrete MMP-2 and MMP-9. Being an inhibitor of MMP-9,TGF-β may be very potential in improving the healing of non-infectious corneal ulcer.
