抗增生药物对人视网膜神经胶质细胞的抑制作用 被引量:1

摘要 目的寻找抑制视网膜、玻璃体内细胞增生的有效药物;明确抗增生药物秋水仙碱、道诺霉素和5-氟尿嘧啶(5-Fu)对体外培养的人视网膜胶质(retinal glia,RG)细胞的作用。方法用 MTT法测定秋水仙碱(0.5~16.0μg/ml)、道诺霉素(0.1~3.2μg/ml)和5-Fu(0.5~16.0μg/ml)对体外培养人RG细胞的作用。结果秋水仙碱(1.0~16.0μg/ml)、道诺霉素(0.2~3.20μg/ml)和 5-Fu(1.0~16.0μg/ml)3组药物对培养细胞均有抑制作用,与对照组均差别显著(P<0.01),ID_(50)分别为3.11μg/ml,0.79μg/ml和5.23μg/ml。结论秋水仙碱、道诺霉素和5-Fu对RG细胞有明显抑制作用。 Objective To select effective drugs against cellular proliferation of cultured human retinal glial cells which might involve in the retinal and/or vitreous proliferation. Methods Human retinal glia cells were cultured by enzyme-digesting method. Colchicine (0. 5 μg/ml, 1. 0 μg/ml,2. 0 μg/ml,4. 0μg/ml, 8.0μg/ml, 16. 0μg/ml), daunomycin (0. 1μg/ml,0. 4μg/ ml,0.8μg/ml, 1. 6μg/ml,3. 2μg/ml) and 5-Fu(0. 5μg/ml, 1. 0μg/ml,2. 0μg/ml,4. 0μg/ml,8.0μg/ml, 16.0μg/ml) were added to the cultures of human retinal glial cells and the proliferative rates of the cells were measured by MTT method. The t test was used for the statistic analysis. Results Colchicine at a dosage ranging from 1.0μg/ml to 16.0μg/ml, daunomycin(0. 2 μg/ml-3.2μg/ml) and 5-Fu(1.0μg/ml-16.0μg/ml) all inhibited the cellular proliferation effectively with their ID_50 of 3.11μg/ml,0.79μg/ml and 5.23μg/ml, respectively. There were significant differences between the test groups and the control groups(P<0.01). Conclusion Colchicine, daunomycin and 5-Fu have an effective inhibition on human retinal glial cell proliferation,which might be the potential candidates for the treatment of proliferative vitreoretinopathy.
