摘要
目的:探讨ATR基因的表达对HeLa细胞对烷化剂敏感性的影响。方法:采用RNA干扰技术抑制HeLa细胞ATR基因的表达,观察ATR被阻断后HeLa细胞对烷化剂敏感性的变化,从而确定ATR基因的作用。结果:筛选到表达针对ATR基因的短发夹RNA(shRNA)阳性克隆,Western印迹结果显示ATR基因表达受到明显抑制;HeLa细胞对烷化剂的敏感性实验提示,ATRshRNA+HeLa细胞对烷化剂的敏感性明显增强。结论:抑制ATR基因可明显提高HeLa细胞对烷化剂的敏感性。
Objective: To investigate the effect of the expression of ATR gene on HeLa cells' susceptibility to al-kylating agents. Methods: RNA interference technology was employed to suppress the expression of ATR, then the role of ATR in HeLa cells' anti-alkylation ability was inspected. Results: A positive clone of siRNA against ATR gene was successfully screened and it can strikingly inhibit the expression of ATR in HeLa cells by Western blot analysis, and then the ability of HeLa cells to resist alkylating agents was significantly attenuated. Conclusion: The inhibition of the expression of ATR gene can significantly improve the HeLa cells' susceptibility to alkylating agents.
