摘要
目的:利用大鼠评价酶解转变的人红细胞的安全性。方法:人红细胞与大鼠血清进行常规配血,检测二者之间的相容性;将人B型红细胞酶解后输注大鼠,流式细胞术检测人红细胞在大鼠体内的存活率。结果:人红细胞与大鼠血清不凝集,但人红细胞输注大鼠体内很快被排斥,1 h下降至50%以下,18 h被完全排斥。结论:仅依靠红细胞配血实验来选择人红细胞输血动物模型是不全面的,大鼠不适合作为酶解转变的人红细胞安全性评价的动物模型。
Objective: To study the safety of enzymatically converted group O red blood cells(ECORBC) by rat model. Methods: The compatibility between human red blood cells(RBC) and rat was detected by blood match- ing. The B group RBC after enzymatic conversion were transfused to rats, and the survival rate of the ECORBC af-ter transfusion was detected. Results: There was no agglutination after human RBC mixed with rat blood serum. ECORBC were rejected by rats within short time, the survival rate of ECORBC decreased below 50% at 1 hour and about zero at 18 hours. Conclusion: Blood matching is not enough to select the animal model for human RBC transfusion, the rat is not suitable to appreciate the ECORBC safety.
