大鼠牙周膜I型胶原mRNA原位杂交方法的建立 被引量:1

摘要 目的 建立检测大鼠牙周膜内 型胶原 m RNA表达的原位杂交方法。方法 选用大鼠为对象建立动物模型 ,采用地高辛标记的寡核苷酸探针的原位杂交方法。结果 阳性对照组 ,用有活跃 型胶原 m RNA表达的大鼠皮肤标本进行原位杂交 ,得到了理想的阳性结果 ;各阴性对照均未检测到信号。标本的信号随着 PK浓度的升高而升高 ,大鼠牙周膜 型胶原 m RNA的表达很强。结论 所建立的原位杂交方法敏感性高 ,特异性高。 WT5”BZ] Objective The aim of this study is to establish method of in situ hybridization for the expression of collagen type Ⅰ in rat periodontal ligament.Methods Male Wistar rats were selected to establish animal model by extracting the left maxillary molars. Oligonucleotide probe labeled with digoxigenin were used in in situ hybridization.Results In positive control group, in situ hybridization was applied to observe the expression of type Ⅰ collagen mRNA in rat skin, ideal positive results were got There were no expression observed in all negative control groups. The strength of the expression increased with the PK concentration increasing. The expression of type Ⅰ collagen mRNA was strong.Conclusion In situ hybridization method established showed sensitivity and specificity. [WT5”HZ]
