摘要
在pH7.4的Tris-HCl缓冲液中,应用紫外-可见光谱法、荧光光谱法、DNA热变性法、盐效应法及黏度法研究硫酸沙丁胺醇(salbutamol sulphate,SS)与鲱鱼精DNA(herring sperm DNA,hsDNA)之间的相互作用.紫外-可见光谱法检测的结果初步排除SS与hsDNA之间存在嵌插作用的可能;DNA热变性法检测的结果表明SS与hsDNA之间可能存在静电或沟槽作用;盐效应法、KI荧光猝灭法及黏度法检测的结果表明SS与hsDNA之间存在静电和沟槽作用;同时研究发现hsDNA使SS的荧光发生静态猝灭,测得两者之间结合常数K=1.51×104 L.mol-1、结合位点数n=1.27.该研究为肾上腺素类药物的开发和不良反应的消除提供了一定的理论依据.
The interaction of salbutamol sulphate(SS) with herring sperm DNA(hsDNA) in the pH7.4 Tris-HC1 buffer solution was investigated by means of LIV-Vis absorption spectra, fluorescence spectra, thermal denaturation of DNA, salt effect and viscosity, respectively. The experimental results from UV-Vis absorption spectra and thermal denaturation of DNA show that the binding mode of SS interacting with hsD- NA might not be in classic intercalation mode but in electrostatic or groove mode. Moreover, the electro- static interaction and groove binding mode was proved by the assay of salt effect,KI fluorescence quenching and viscosity. The fluorescence quenching model between SS and hsDNA belonged to static fluorescence quenching. The binding constant for SS with hsDNA was K= 1.51 × 104 L mol-l , and the number of binding sites was n= 1.27. This work may be usefully applied to developing the adrenergic agents and elimi- nating the adverse reactions.
