摘要 目的 探讨高密度脂蛋白(HDL)的体外氧化过程及丙丁酚对HDL氧化的抑制作用。方法 采用一次性密度梯度超速离心法获取人血清HDL,以CU~2+介导其氧化,荧光法检测样品中硫代巴比妥酸反应产物(TBARS)的含量,用于判定氧化效果。结果Cu2+可有效地介导HDL的体外氧化,并在一定范围内呈浓度和时间依赖性;预先加入丙丁酚可有效阻止HDL的体外氧化,5μmol/L丙丁酚即有显著的抗氧化作用。结论 HDL亦易于发生氧化修饰,抗氧化剂丙丁酚可明显抑制由Cu2+介导的HDL体外氧化。 Objective To investigate the oxidation of high-density lipoproteins(HDL) in vitro and protective effects of probucol. Methods Human plasma lipoproteins were separated by one-step ultracentrifugation with a simple density-gradient. The in vitro oxidation of HDL was mediate by CuSO4, the oxidative effect of Cu2+ and the antioxidation effect of probucol were assayed simultaneously by thiobarbituric acid reactive substances (TBARS). Results (1) Cu2+ could effectively mediated the oxidation of HDL in vitro, which was both dose-dependent and time-dependent with Cu2+ within definite ranges.(2) The oxidation of HDL in vitro could be inhibited by probucol, and 5μmol/L probucol was effective obviously. Conclusion HDL is susceptible to oxidation and probucol can inhibit the Cu2+ mediated oxidation of HDL in vitro.
