摘要
目的:研究类风湿关节炎(Rheumatoid arthritis,RA)患者外周血、滑液和滑膜组织中趋化因子CCL28的表达。方法:酶联免疫吸附试验(ELISA)法检测正常人血浆、RA患者血浆及滑液CCL28水平,免疫组织化学法分析RA和骨关节炎患者滑膜组织中CCL28表达。结果:活动期RA患者有关节积液组血浆CCL28水平高于无关节积液组(P<0.01)和健康对照组(P<0.05),其滑液CCL28水平显著高于配对血浆(P<0.001);CCL28水平与RA患者的临床和实验室指标无关。RA滑膜组织中,CCL28表达于衬里层和衬里下层的巨噬细胞及中性粒细胞,而骨关节炎对照滑膜几乎无CCL28表达。结论:RA患者炎症关节中CCL28的表达上调,CCL28可能是介导炎性细胞迁移进入关节的重要趋化因子。
Objective:To examine the expression of the CC chemokine CCL28 in peripheral blood,synovial fluid and synovial tissue of patients with rheumatoid arthritis(RA).Methods: CCL28 levels in normal plasma,RA plasma and synovial fluid were measured by enzyme-linked immunosorbent assay(ELISA).CCL28 expression in synovial tissue from RA and osteoarthritis patients was analysed by immunohistochemistry.Results: Active RA patients with joint effusion had higher plasma CCL28 levels than those without joint effusion(P0.01) and healthy controls(P0.05).CCL28 levels in synovial fluid were significantly higher than that in paired plasma(P0.001).CCL28 levels were not related to clinical features of RA patients.CCL28 expression was observed in macrophages and neutrophils of the intimal lining and sublining regions in RA synovium,but was hardly detectable in osteoarthritis synovial tissues.Conclusion: The expression of CCL28 is up-regulated in inflamed joints of RA patients and CCL28 might be an important chemokine involved in migration of inflammatory cells into the joint.
