摘要 目的:探讨长波紫外线(Ultraviolet A,UVA)辐射对人体成纤维细胞形态、增殖活性的影响及作用机制。方法:将人体成纤维细胞分为空白对照组、低(1 J/cm2)、中(5 J/cm2)及高(10 J/cm2)UVA辐射组。辐射后6、24、48、72小时观察细胞形态、增殖活性及IL-1α、IL-1β、IL-6的水平。结果:中、高剂量UVA辐射后6小时细胞形态明显改变,并分别于48和24小时细胞增殖活性最低,IL-1α、IL-1β、IL-6的水平于辐射后6小时显著升高,且IL-1β、IL-6持续高水平至48~72小时。结论:中、高剂量UVA辐射对人体成纤维细胞有明显的光损伤作用,其机制可能与UVA刺激成纤维细胞分泌的炎性细胞因子有关。 Objective:To investigate the effects of ultraviolet A(UVA) on the morphology and proliferation activity of human fibroblasts and the mechanism of action.Methods: Human fibroblast were respectively divided into blank group and low(1 J/cm2),moderate(5 J/cm2),high(10 J/cm2) UVA groups.The alterations of cellular morphology and proliferation,as well as the secretion of inflammatory cytokines were measured 6,24,48,72 h after irradiation.Results: Obvious changes of cellular morphology were observed as early as 6 h after moderate and high UVA irradiation.At the same time,the level of IL-1α,IL-1β,IL-6 proteins were significantly higher than blank control,and the high level of IL-1β and IL-6 maintained to 48-72 h.The cell proliferation decreased to the lowest level 48 h after moderate UVA irradiation and 24 h after high UVA irradiation,respectively.Conclusion: Moderate and high dose UVA irradiation have significant light-damage on human fibroblast,and the mechanism may be related with inflammatory cytokines,which secreted by UVA stimulated fibroblast cells.
