摘要
目的:探讨花旗松素和槲皮素对细菌脂多糖(LPS)诱导流产小鼠子宫巨噬细胞的免疫抑制作用。方法:昆明种正常孕鼠90只,随机分为对照组(A组);实验组均于孕6天尾静脉注射LPS 0.2 ml诱导流产小鼠,并分为LPS处理组(B组),低浓度花旗松素及LPS双处理组(C1组)、高浓度花旗松素及LPS双处理组(C2组)、低浓度槲皮素及LPS双处理组(D1组),高浓度槲皮素及LPS双处理组(D2组),每组15只。用酶组织化学、免疫组化和ELISA方法检测孕9天子宫角组织非特异性酯酶阳性(α-NAE+)巨噬细胞,CD14+巨噬细胞和TNF-α含量。结果:B组小鼠流产率为100%,胚胎吸收率为100%(P<0.01);灌胃花旗松素和槲皮素可以显著抑制流产模型小鼠的流产率和胚胎吸收率(P<0.01)。B组孕鼠子宫内膜和肌层α-NAE+,CD14+巨噬细胞数量和阳性面积与A组相比均显著增多(P<0.01);各双处理组与B组相比,子宫内膜和肌层α-NAE+巨噬细胞和CD14+巨噬细胞数目和阳性面积显著减少(P<0.01)。B组孕鼠子宫TNF-α含量与A组相比显著增加(P<0.05),而双处理组接近正常水平。结论:通过抑制子宫巨噬细胞的数量、分布、活性和TNF-α分泌量,可能是花旗松素和槲皮素对抗LPS诱导小鼠流产的机理之一。
Objective:To explore the immunosuppression effects of taxifolin and quercetin on the macrophages in the uterine of abortion mice induced by LPS.Methods: Gravidity mice of Kunming(90 mice) were divided into control group(group A)and experimental group.In order to induce abortion,mice in experimental group were intravenous injected LPS on 6 day of pregnancy.Experimental group were divided into group B,group C1(ingestion low concentration taxifolin),group C2(ingestion high concentration taxifolin),group D1(ingestion low concentration quercetin) and group D2(ingestion high concentration quercetin),each group of 15 mice.Then the variety of α-NAE+,CD14+ macrophages and TNF-α in the uteru of gravidity mice were identified by enzyme-histochemistry,immunohistochemistry and ELISA on 9 day of pregnancy.Results: The abortion rate of gravidity mice in group B was 100%,the fetal resorbing rate was 100%(P0.01).The abortion rate and the fetal resorbing rate of gravidity mice decreased distinctly through oral taxifolin or quercetin(P0.01).The quantities and positive cell area of α-NAE+macrophages and CD14+macrophages in the endometrium and the myometrium of gravidity mice of group B were greatly increased compared with group A(P0.01).The α-NAE+ macrophages and CD14+ macrophages quantities and positive cell area in the endometrium and the myometrium of the taxifolin and quercetin treatment group were reduced compared with group B(P0.01).The TNF-α contents in the uteri of mice in group B were greatly increased compared with group A(P0.05),but close to normal levels in the taxifolin and quercetin treatment group.Conclusion: The quantity,distribution and activity of macrophages and the secretion TNF-α were inhibited,which maybe one of the mechanisms for the miscarriage induced by LPS could be antagonized by taxifolin and quercetin.
