摘要
目的 研究胰头癌组织与正常胰腺组织中γ 谷氨酰转肽酶 (γ glutamyltranspeptidase,GGT)基因的表达及核苷酸序列的差异 ,以阐明胰头癌患者血清GGT异常升高的分子机制。方法 收集 3例胰头癌、2例胆管癌和 2例正常胰腺的手术切除标本。应用逆转录多聚酶链反应及核苷酸序列分析方法 ,检测不同组织中GGTmRNA表达量的变化及GGT基因核苷酸序列。结果 胰头癌与正常胰腺组织中的GGTcDNA核苷酸序列中 12 0 2bp~ 16 0 5bp相一致。在正常胰腺、胆管癌和胰头癌组织中均可见GGTmRNA的表达扩增产物 ,但胰头癌组织中GGT基因表达水平明显高于胆管癌和正常胰腺组织。结论 胰头癌与正常胰腺组织中的GGT基因序列是相同的 ,但胰头癌组织中GGTmRNA表达量明显增高 ,证实GGT是诊断胰头癌的有效指标。
Objective[WT5”BZ] To explore the molecular mechanism of the elevation of serum γ-glutamyl transpeptidase(GGT) activity in pancreatic head cancer.[WT5”HZ] Methods [WT5”BZ] GGT mRNA expression was measured by reverse transcription-polymerase chain reaction(RT-PCR) and GGT gene sequence analysis was made from sugically removed tumor tissues of pancreatic head cancer (3 cases),cholangiocarcinoma (2 cases).Results were compared with those from normal pancreatic tissue (2 cases).[WT5”HZ]Results [WT5”BZ] The nucleotide sequence of GGT cDNA fragment from pancreatic head cancer tissues was coincidental with that from normal human pancreas with range of 1*!202*!bp~1605*!bp. GGT mRNA were detected in all tissues from normal human pancreas, cholangiocarcinoma and pancreatic head cancer, but the expression level of GGT mRNA in three cases of pancreatic head cancer is significantly higher than that in cholangiocarcinoma and normal pancreas. [WT5”HZ]Conclusion[WT5”BZ] The GGT cDNA gene sequence in pancreatic head cancer is same as in normal pancreas. But the expression level of GGT mRNA increased significantly in cases of pancreatic cancer. The higher expression of GGT in pancreatic head cancer tissues indicated that it could be a potent diagnostic index. [WT5”HZ]
