The expression of cyclin D1,CDK4,p16, Rb and their significance in benign and malignant lesions of the pancreas

摘要 目的 研究胰腺良性和恶性病变组织中cyclinD1、CDK4、p16和Rb的表达及意义。方法 对 5 3例胰腺癌、17例慢性胰腺炎石蜡包埋切片行ABC免疫组织化学法染色。结果 慢性胰腺炎和癌旁上皮组织的cyclinD1、CDK4、Rb阳性率明显低于胰腺癌 ,而p16则相反 ;cyclinD1、CDK4和Rb阳性及p16阴性的慢性胰腺炎和癌旁上皮多为上皮中度至重度不典型增生 ;高分化和未转移胰腺癌cyclinD1、CDK4和Rb阳性率明显低于低分化和有转移的胰腺癌 ,而p16则相反 ;cyclinD1表达与CDK4表达呈密切正相关 ,p16表达与Rb表达呈密切负相关 ,cyclinD1、CDK4表达与Rb表达呈正相关趋势 ,与p16表达呈负相关趋势。结论 cyclinD1、CDK4、p16和Rb表达特征与胰腺癌的发生发展、生物学行为及其预后有密切关系。 Objectives[WT5”BZ] To investigate the expression of cyclin D1,CDK4, p16,Rb and their significance in benign and malignant lesions of the pancreas.[WT5”HZ] Method[WT5”BZ] Avidin-biotin complex method was used on paraffin-embedded section to investigate tissues of pancreatic carcinoma in 53 cases and 17 cases with pancreatitis.[WT5”HZ] Results[WT5”BZ] The positive rates of cyclin D1,CDK4 and Rb were significantly higher in cases with pancreatic carcinoma than that with chronic pancreatitis and paracancerous epithelium, the expression of p16 was lower in the former than that in the later. Most of the positive cases of cyclin D1, CDK4, Rb and the negative ones of p16 were mildly or severely atypical hyperplasia of pancreatic epithelium in chronic pancreatitis and paracancerous epithelium. The positive rates of cyclin D1, CDK4 and Rb in the pancreatic carcinoma of well-differentiation and without metastasis[CM(43] were significantly lower than that in the ones of poor-differentiation and with metastasis, and that of p16 was the opposite. The positive correlation was found between the expressison of cyclin D1 and CDK4, the negative correlation was also found between the expressison of p16 and Rb in pancreatic carcinoma.[WT5”HZ] Conclusions[WT5”BZ] The expressions of cyclin D1, CDK4, p16 and Rb might be related to the carcinogesis, development, biological behaviors and prognosis of pancreatic carcinoma. Abnormality in the negative feedback regulatory pathway of cyclin D1/CDK4, Rb and p16 might be involved in the molecular mechanism of pancreatic carcinoma. [WT5”HZ]
