利用噬菌体肽库技术进行抗原中和位点的鉴定

摘要 目的确定病毒 IBDV(infection bursal diseases virus, IBDV)的中和位点,为生产病毒疫苗奠定基础。方法利用噬菌体肽库技术,模拟病毒的病原决定簇,同抗IBDV的单抗相结合,通过对肽库的筛选和测序,从而确定病毒的中和位点的氨基酸序列。结果与IBDV抗原的原序列进行对应查找,发现原序列上的198~208(Asp-Ser-Asp-Arg-Pro-Arg-Val-Tyr-Thr-Ile)这11个氨基酸能够体现中和位点的特点,并且与所测序列有4~5个氨基酸残基的重复。结论该序列就是抗原的中和位点。 Objective To locate the neutralizing epitope of virus(IBDV), it is the basis of virus vaccine. Methods Phage peptide library as a epitope of virus were combined to anti-McAB of IBDV, through screening and sequencing, amino acid sequencies of virus epitope were determined. Results By comparing these amino acid with IBDV,the neutralizing site of virus were located at the residue from 198 -208 (Asp- Ser- Ser- Asp-Arg- Pro- Asg- Val-Tyr-Thr-Ile). These 11 amino had the characteristic of neutralizing epitope and repeat the amino sequencing for 4 - 5 amino residue. Conclusion This sequences is the neutralizing epitope.
