摘要
[目的]探讨太白楤木对肝纤维化模型大鼠Ⅰ型胶原纤维(Col-Ⅰ)、Ⅲ型胶原纤维(Col-Ⅲ)、α-平滑肌肌动蛋白(α-SMA)表达的干预作用。[方法]雄性SD大鼠60只,随机分为阴性对照组、模型组、太白楤木大、中、小剂量组和秋水仙碱组,采用40%CCl4橄榄油腹腔注射诱导肝纤维化模型,2次/周,连续8周。采用苏木精-伊红及Mas-son染色观察肝组织病理学改变;RT-PCR检测Col-Ⅰ、Col-ⅢmRNA表达水平;Western-blot检测α-SMA的表达水平。[结果]与模型组相比,太白楤木大、中、小剂量组及秋水仙碱组肝细胞变性坏死减轻,纤维条索减少,且秋水仙碱组、太白楤木小、中、大剂量组呈逐渐减轻趋势。与模型组相比,太白楤木大、中、小剂量组及秋水仙碱组Col-Ⅰ、III mRNA表达水平均减低(均P<0.05);与秋水仙碱组比较,太白楤木大剂量组Col-Ⅰ、III mRNA表达减低(均P<0.05)。太白楤木大、中、小剂量组和秋水仙碱与模型组相比,a-SMA表达不同程度减低(均P<0.05),与秋水仙碱组相比,太白楤木大、中剂量组a-SMA表达均减低(均P<0.05)。[结论]太白楤木可通过抑制肝组织中胶原纤维的增生和肝星状细胞的活化,从而减轻肝纤维化。
[Objective]To explore the effect of Aralia taibaiensis on expression of Col-Ⅰ,Col-Ⅲ and a-SMA in hepatic fibrosis rats.[Methods]Sixty male Sprague-Dawlley(SD) rats were randomly divided into six groups: negative control group,model group,high-dose,middle-dose,low-dose of Aralia taibaiensis groups and colchicine group.Hepatic fibrosis models were induced by CCl4.The change of liver histopathology was observed by HE and Masson staining.The mRNA of Col-Ⅰand Col-Ⅲ were detected by RT-PCR.The protein of α-SMA was detected by western blot.[Results]1.Compared with model group,the fiber hyperplasia and hepatocellular necrosis were significantly decreased in Aralia taibaiensis treated groups and colchicine group.In addition,the degree of liver fibrosis was gradually relieved following the increasing doses of Aralia taibaiensis.2.Compared with model group,the mRNA level of type I and III collagen fiber was significantly decreased in Aralia taibaiensis groups and colchicine group(all P 0.05).3.Compared with colchicine group,the mRNA level of type I and III collagen fiber was significantly decreased in Aralia taibaiensis high-dose group(P0.05).Compared to model group,α-SMA expression in Aralia taibaiensis groups and colchicine group was reduced with different degrees(all P 0.05).Compared with colchicine group,the protein level of α-SMA expression was significantly reduced in Aralia taibaiensis high and middle-dose groups(P 0.05).[Conclusion]Aralia taibaiensis can improve liver fibrosis via inhibiting the expression of Col-Ⅰ,Col-Ⅲ and a-SMA expression.
