白藜芦醇对糖尿病下肢缺血大鼠微血管新生的影响 被引量:3

摘要 目的探讨白藜芦醇对糖尿病下肢缺血大鼠缺血下肢骨骼肌中缺氧诱导因子-1α(HIF-1α)及血管内皮生长因子(VEGF)表达水平的影响,以及刺激侧支微血管新生的作用。方法将40只SD大鼠随机分为5组:假手术组、下肢缺血组、糖尿病组、糖尿病下肢缺血组及白藜芦醇治疗组。假手术组仅行假手术,不建立模型;下肢缺血组仅建立下肢缺血模型;糖尿病组仅建立糖尿病模型;糖尿病下肢缺血组先建立糖尿病模型,再建立下肢缺血模型;白藜芦醇治疗组按糖尿病下肢缺血组方法建立糖尿病下肢缺血模型后,以白藜芦醇按20 mg/(kg.d)剂量连续腹腔注射14 d。HE染色测定左下肢骨骼肌中毛细血管密度(MVD),Western blot检测各组HIF-1α蛋白表达量,免疫组化检测各组VEGF蛋白表达量。结果下肢缺血组MVD及HIF-1α、VEGF蛋白表达量均较假手术组明显升高(P<0.01),糖尿病下肢缺血组MVD及HIF-1α、VEGF蛋白表达量均较下肢缺血组明显降低(P<0.01),而白藜芦醇治疗组MVD及HIF-1α、VEGF蛋白表达量均较糖尿病下肢缺血组明显升高(P<0.05)。结论白藜芦醇治疗可提高糖尿病下肢缺血状态下HIF-1α及VEGF蛋白等促血管生成因子的表达量,有利于微血管新生,改善局部循环。
