原发性肝癌中MMP-9、MMP-13和TIMP-3的表达及意义 被引量:7

The expression of MMP-9,MMP-13,and TIMP-3 in hepatocellular carcinoma

摘要 目的探讨基质金属蛋白酶(MMP)-9、MMP-13与组织金属蛋白酶抑制剂(TIMP)-3在肝癌中的表达及与肝癌发生、发展的关系。方法通过免疫组织化学检测80例肝癌组织及60例非肿瘤肝组织中MMP-9、MMP-13和TIMP-3的表达,并分析其与肿瘤发生、大小、转移等的关系。结果与正常肝组织比较,肝癌组织MMP-13阳性表达率及TIMP-3阴性表达率均显著升高(91.2%vs 75.0%及60.0%vs 38.3%,P<0.05)。大肝癌(最大径≥5 cm)组织中MMP-9的阳性表达率显著高于小肝癌(最大径<5 cm)(100.0%vs 83.3%,P<0.05);发生转移的肝癌组织中MMP-13的阳性表达率显著高于未转移者(97.7%vs 83.8%,P<0.05)。而低分化、转移与肝癌组织中TIMP-3阴性表达独立相关(P<0.05)。结论 MMP-13阳性表达及TIMP-3的阴性表达与肝癌的发生相关,而MMP-13、MMP-9的阳性表达及TIMP-3的阴性表达与肝癌的发展和转移有关。 Objective To investigate the roles of MMP-9,MMP-13,and TIMP-3 in the pathogenesis and metastasis of hepatocellular carcinoma(HCC).Methods With immunohistochemistry,MMP-9,MMP-13,and TIMP-3 were detected in 80 HCC tissues and 60 normal liver tissues.The correlations of MMP-9,MMP-13 and TIMP-3 with the tumor size,histological grade,and metastasis status were evaluated.Results Compared with normal liver tissues,the MMP-13 positive rate and TIMP-3 negative expression rate in HCC tissues were significantly increased(91.2% vs 75.0% and 40.0% vs 61.7%,P〈0.05).Significantly higher MMP-9 positive rate was observed in HCC with big size(DIA≥5 cm,100.0% vs 83.3%,P〈0.05);so was significantly higher MMP-13 positive rate in HCC with metastasis(97.7% vs 83.8%,P〈0.05).Conclusion The missing expression of TIMP-3 in HCC was significantexpression of MMP-13 and MMP-9,and missing expression of TIMP-3 are correlated with the progression and metastasis of HCC.
