鼻息肉调节激活正常T细胞表达和分泌因子的测定及其意义 被引量:16

摘要 目的 探讨在鼻息肉形成过程中 ,上皮应答时产生调节激活正常T细胞表达和分泌因子 (regulateduponactivation ,normalTcellexpressedandsecreted ,RANTES)对嗜酸粒细胞趋化、移行、局部聚集的影响。方法 采用无血清原代细胞培养法培养鼾症患者下鼻甲上皮细胞和鼻息肉上皮细胞 ,经炎性介质IL 1β(2 5 μg/L ,5 0 μg/L)刺激后收集 2 4h和 48h上清液 ,以酶联免疫吸附试验 (enzyme linkedimmunosorbentassay,ELISA)法测定上清液中RANTES的浓度。结果 ①鼾症患者下鼻甲上皮细胞经IL 1β刺激后上清液中RANTES的平均水平比刺激前约提高 1 5~ 10倍 ;鼻息肉上皮细胞刺激后RANTES的平均水平比刺激前约提高 10~ 2 0倍 ,2组刺激前后差异均有显著性 (t检验 ,P <0 0 0 1)。②刺激前鼻息肉上皮细胞中RANTES的水平与下鼻甲上皮细胞中的水平差异无显著性 (t检验 ,P >0 0 5 ) ,但刺激后 ,对应于IL 1β作用的不同浓度 ,不同时间RANTES在鼻息肉上皮细胞中的水平均显著高于下鼻甲上皮细胞中的浓度 (t检验 ,P <0 0 0 1)。③IL 1β作用 48h ,2组RANTES的水平均随着IL 1β刺激浓度的加大而增高 ;在一定的刺激浓度下 ,2组RANTES的水平随着刺激时间的延长而增加。结论 人鼻粘膜上皮细胞能够产生趋化因子RANTES ,它是鼻息? Objective To investigate the effect of RANTES(regulated upon activation,normal T cell expressed and secreted)on chemotatis, migration and accumulation of eosinophils in nasal polyps during epithelial immune responses Methods The epithelial cells were obtained from nasal polyps and inferior turbinates in patients with snoring , and cultured for 24 and 48 hours with IL-1β(25 μg/L,50 μg/L) RANTES was measured in the culture supernatant by ELISA Results ① On incubation with IL-1β, the epithelial cells from inferior turbinate and nasal polyps released 1 5-10 fold and 10-20 fold greater amounts of RANTES respectively than unstimulated samples Significant increase in levels of RANTES was found in both groups( P <0 001); ②There was no difference between nasal polyp and inferior turbinate before incubation with IL-1β( P >0 05) in the level of RANTES After IL-1β stimulation, the concentration of RANTES was higher in polyp than in inferior turbinate ( P <0 001); ③ The expression of RANTES in both groups was dose-dependent and time-dependent upon stimulation of IL-1β Conclusion The epithelial cells of nasal mucosa are active end-organs, capable of releasing chemokine RANTES In nasal polyps, RANTES has chemotactic activity for eosinophils and other inflammatory cells It could affect the chemotaxis and eosinophil function and may play an important role in polyp formation.
