摘要
目的 :观察不同巯基蛋白酶抑制剂对不同恶性度胃癌细胞株体外侵袭转移力的影响。方法 :用体外粘附法和 Boyden小室模型法观察外源性巯基蛋白酶抑制剂 E- 6 4和内源性抑制剂胱蛋白 A(Cystatin A)对高分化胃腺癌 MKN2 8细胞株和低分化胃腺癌 MKN45细胞株细胞体外的侵袭转移能力。结果 :E- 6 4和Cystatin A抑制二胃癌细胞株体外的细胞粘附力和细胞对基质胶 (matrigel)的侵袭力 ,对 MKN2 8细胞的作用较 MKN45细胞明显。结论 :巯基蛋白酶 Cathepsin B参与胃癌的侵袭转移过程 。
Objective:To observe the effect of cysteine proteinase inhibitors(E 64 and cystatinA) on human gastric carcinoma cell lines(MKN 45 and MKN28) in terms of invasion and metastasis.Methods:To examine the cancer cell ability to interact with extracellular matrix components(laminin and fibronectin),and the ability to invasion to matrigel and motility in Boyden chamber.Results:E 64 and Cystatin A inhibit adherence of gastric cancer cells on laminin or fibronectin, also inhibit invasion of gastric cancer cells to metrigel, no effect on motility of cancer cells.Conclusion:Cysteine proteinase and cysteine proteinase inhibitor play an important role in the invasion and metastasis of gastric cancer cells.[FK(WB80011。6]
