摘要
分子诊断是分子病理研究的最主要内容和手段 ,对肿瘤病理学研究由形态学深入到分子水平具有十分重要的意义。本文就分子诊断在肿瘤研究中的意义和应用 ,基因过表达和基因突变的检测、等位基因分析、微卫星不稳定性分析、端粒酶检测等方面技术方法和应用作一介绍 ,包括PCR技术、荧光原位杂交 (FISH)、比较基因组杂交 ,寡核苷酸引物原位DNA合成法(PRINS)、DNA芯片技术等 ;并对分子诊断发展前景及标准化问题进行了讨论。
Molecular diagnosis has important significance for development of tumor pathology from morphological to molecular level as it is the main contents and methods of molecular pathology.This paper has reviewed the application and significance of molecular diagnosis in tumor researches,include detection of gene over-expression and gene mutation,analysis of allelic gene,analysis of MI,detection of telomorase,and their associated technique include PCR,FISH,PRINS,comparative hybridization of genome and DNA chip technique,and also discussed developing prospect of molecular diagnosis and its standardization.
