呼吸道合胞病毒对人肺腺癌细胞系PAa的影响

Effects of respiratory syncytial virus on human lung adenocarcinoma cell line PAa and p53 gene mutation

目的 :探讨呼吸道合胞病毒 (RSV)感染对体外培养人肺腺癌细胞系PAa的影响。方法 :应用RSV体外感染PAa细胞 ,观察PAa细胞的变化 ;聚合酶链反应 (PCR)结合单链构象多态性分析 (SSCP)检测PAa细胞 p5 3基因突变情况。 结果 :RSV感染PAa细胞 2 4h后 ,细胞出现融合病变 ,形成巨融合细胞 ;病变细胞p5 3第 5、6、7、8、9外显子突变热点区域未见单链多态性变化。结论 :RSV对PAa细胞形态影响明显 ,对PAa细胞基因水平的改变有待深入研究。

Objective: To study the effects of respiratory syncytial virus (RSV) on the morphology of human lung adenocarcinoma cell line PAa and the mutation of p53 Methods:PAa cells were infected with RSV and the mutation of p53 in PAa cells was observed with polymerase chain reaction single strand conformation polymorphism (PCR SSCP) Results: 24 hours after RSV infection, huge fused PAa cells were formed PCR SSCP revealed no change of exon 5, 6, 7, 8 and 9 of p53 gene in the fused PAa cells Conclusion: RSV infection affects significantly the morphology of PAa cells but its effects on the changes of genes in PAa cells needs further investigation
