摘要
目的 :探讨低浓度CO对大鼠主动脉平滑肌细胞迁移的影响。方法 :体外培养大鼠主动脉平滑肌细胞 ,用低浓度CO处理 2 4、48、72h ,检测平滑肌细胞迁移情况。结果 :2 4、48、72h后未加处理的对照组大鼠主动脉平滑肌细胞迁移距离逐渐增加 ,与 2 4h组比差异显著 (P <0 0 0 1)。 2 0× 10 -6、5 0× 10 -6、80× 10 -6CO处理细胞 2 4h后 ,细胞迁移距离逐渐下降 ,2 0× 10 -6CO组与对照组差异不显著 (P <0 0 5 ) ,其余两组均显著低于对照组 (P <0 0 1)。 2 0× 10 -6、5 0× 10 -6、80× 10 -6CO处理细胞 48、72h后 ,其迁移距离也显著低于对照组 (P <0 0 1)。结论 :大鼠主动脉平滑肌细胞自身存在迁移情况 ,培养时间愈长 ,迁移距离愈大。低浓度CO可以显著抑制平滑肌细胞的迁移 ,随着浓度增加 ,抑制作用增强。
Objective: To investigate the effects of exogenous carbon monoxide (CO) on the migration of cultured vascular smooth muscle cells (SMCs) Methods: Pulmonary artery SMCs were cultured and then treated with exogenous CO for 24, 28 and 72 h respectively The migration distances of the SMCs were determined by Sarkar′s method Results: The migration distances of the SMCs treated for 24,28 and 72 h were increased gradually (24 h group vs control:P< 0 001) The migration distances of the SMCs were decreased after 24 h of the treatment with 20×10 -6 , 50×10 -6 , 80×10 -6 CO (20×10 -6 CO group vs control: P< 0 05; 50×10 -6 ,80×10 -6 CO group vs control: P< 0 01) After 72 h of the treatment, the distances were significantly lower than those in the control (P< 0 01) Conclusion:[WT5,5”BZ] SMCs may migrate under the condition of normal culture The migration distance is increased with prolongation of culture CO of low concentration can inhibit the migration of SMCs in a concentration dependent manner
