烧伤后肠系膜淋巴结GP130基因表达与细胞凋亡

Expression of GP130 in mesenteric lymph node and its relationship with cell apoptosis after burn injury in mice

目的 观察烧伤后肠系膜淋巴结GP130基因表达与细胞凋亡变化的时相关系。方法 BALB c种小鼠 60只 ,分为致伤组、正常对照组。致伤组动物予以 2 0 %TBSAⅢ度烧伤。于伤后 1、2、3d活杀致伤组 ,并活杀对照组。取肠系膜淋巴结 :ELISA检测IL 6水平 ;提取RNA ,RT PCR检测GP130基因表达变化 ;采用TUNNEL法对细胞凋亡进行观察。结果 在伤后 3d以内IL 6含量明显升高 ;GP130基因表达、细胞凋亡在伤后 1d明显增加。结论 在烧伤后早期 ,肠系膜淋巴结IL 6、GP130表达与细胞凋亡均增加。

Objective To study the change of IL 6R GP130 gene expression in mesenteric lymph nodes (MLN) after burns and its relationship with cell apoptosis. Methods Sixty mice were devided into control group and injury group .In the injury group, animals were inflicted with 20% TBSA full thickness burns. On 1 d,2 d and 3 d after burns, mice were killed, and the mesenteric lymph nodes were taken. IL 6 were measured with ELISA. Isolated cells from the mesenteric lymph nodes were examined with RT PCR to observe the expression of GP130 gene. Meanwhile cell apoptosis was investigated with “TUNNEL” staining. Results IL 6 was increased on 3 d after burns, Expression of GP130 and cell apoptosis were increased on 1 d after burns. Conclusion In the early period postburn,expression of IL 6 and GP130 are increased, so does cell apoptosis.
