摘要
目的:通过检测冠状动脉粥样硬化患者外周血单个核细胞(PBMC)中单磷酸腺苷激活的蛋白激酶(AMPK)以及沉默信息调节因子2相关酶I(SIRT1)的基因表达情况,初步探讨AMPK和SIRT1基因表达与冠状动脉粥样硬化形成以及调控血浆中一氧化氮(NO)水平、血脂、血糖代谢的可能机制。方法:利用普通PCR、琼脂糖凝胶电泳、实时荧光定量PCR及SPSS分析软件检测冠心病者和正常对照组外周血单个核细胞(PBMC)中AMPK和SIRT1的基因表达区别,以及它们与血脂(TC、TG、LDL-c、HDL-c)、空腹血糖(FBG)、一氧化氮(NO)含量的相关性。结果:冠心病组PBMC中AMPK和SIRT1基因表达以及血浆中NO含量均明显低于对照组,差异具有统计学意义(P均<0.05)。AMPK和SIRT1表达均与TC、TG、LDL-c、FBG呈负相关,与NO含量及HDL-c呈正相关(P均<0.05)。结论:冠心病患者PBMC中AMPK和SIRT1基因表达下降可能与冠心病的发病存在相关性,对调控血脂、血糖及NO生成释放中可能起到一定作用。
Objective:To explore the adenosine monophosphate activated protein kinase(AMPK) and silent information regulator 1(SIRT1)gene expression involved in the progression of coronary heart disease,and the possible metabolic mechanism of controlling the serum nitric oxide(NO),lipid and glucose levels in patients with coronary artery disease patients through the detection of gene expression of(AMPK)and(SIRT1)of the peripheral blood mononuclear cell(PBMC).Methods:Detecting the differences in the PBMC genetic expression of AMPK and SIRT1between disease group and control group by ordinary PCR,agarose electrophoresis,and real-time fluorescence quantification methods.Statistical analysis was performed with SPSS 18.0software.Correlation between AMPK,SIRT1gene expression and the other atherosclerosis related factors including lipid,fasting blood glucose,nitric oxide were analyzed.Results:Gene expression of AMPK,SIRT1and serum nitic oxide concentration of the disease group was significantly decreased compared with the control group(Pall0.05).AMPK and SIRT1gene expression were negatively correlated with TC、TG、LDL-c、and FBG,while positively correlated with NO,HDL-c(P all0.05).Conclusions:Decreased expression of AMPK and SIRT1gene are correlated with the occurrence of the coronary heart disease indicating they play a role in controlling blood lipid,glucose and the generation of nitric oxide.
