摘要
目的:建立一种基于大分子物质环糊精的共振散射光色谱差异测定牛奶中残余黄曲霉毒素M l的新型检测方法。方法:β-环糊精与黄曲霉毒素M l结合后,不同浓度黄曲霉毒素M l与环糊精形成的包合物存在不同强度的共振散射光光谱,用共振瑞利散射光对黄曲霉毒素M l进行检测。结果:在波长为335 nm处共振光散射增强强度与黄曲霉毒素M l的浓度呈线性关系。在pH=4.0~5.0时,包合物形成的最佳温度为60℃,静置时间为1 h,方法的检出限为0.38 ng/ml;线性范围为0 ng/ml^8.0 ng/ml,线性相关系数r=0.992回收率为85%~108%。结论:本方法可简便,快速地测定牛奶中的黄曲霉毒素M l,结果可靠。
objective: A new method for determination of residual aspergillus flavus toxin M1 in milk was estab-lished based on macromoleeular material cyclodextrin and resonance light scattering spectrum difference. Methods: After combination of beta cyelodextrin and aspergillus flavus toxin M1, clathrate formed by different concentrations of aspergillus flavus toxin M1 and cyclodextrin showed resonance scattering light spectrum had various intensity, then resonance rayleigh scattering light was used to detect aspergillus flavus toxin M1. Results: At the wavelength of 335 nm, the intensity of resonance light scattering was linear with concentration of aspergillus flavus toxin M1. When the pH value was 4.0 - 5.0, the optimum temperature of inclusion formation was 60 ℃ and the standing time was 1 h. The detection limit of the method was 0.38 ng/ml, the linear range was 0 ng/ml - 8.0 ng/ml with correlation coefficient of r = 0. 992 and recovery of 85 % - 108%. Conclusion: The method is simple and fast in determining as- pergillus flavus toxin M1 in milk, with reliable results.
