摘要
目的:建立了离子色谱法测定全血样品中胆碱的检测方法。方法:对全血样品中胆碱的提取方法和仪器条件进行了优化。烘干磨碎后的血样经曲拉通X-100分散后,加入盐酸溶液在70℃水浴下水解,稀释后样液过on guard RP小柱过滤,采用阳离子色谱柱(IonPac CS12,4 mm)分离,淋洗液为16 mmol/L的甲烷磺酸溶液等度洗脱。结果:在0.05 mg/L^2.0 mg/L质量浓度范围内,胆碱工作曲线的线性相关系数r2为0.999。该方法的检出限为10.0 mg/kg,定量限为30.0 mg/kg。对样品进行加标,胆碱的回收率为95.7%~110.3%。结论:该方法方便可靠,可应用于全血样品中胆碱的检测。
Objective: A method for determination of choline in blood by ion chromatography was established. Methods : Samples were dispersed by Triton X - 100 solution, hydrolyzed by hydrochloric acid in 70℃ water bath, diluted with water and filtered through RP column, then separated on IonPac CS12 cation exchange column with 16 mmol/L methanesulfonic acid solution for gradient elution. Results : In the concentration range of 0.02 mg/L - 5. 0 mg/L, the working curve linear correlation coefficient for choline was r2 =0. 999. The detection limit was 10.0 mg/kg, the limit of quantification was 30.0 mg/kg. The recovery was 95.7% - 110.3%. Conclusion: This method is convenient and reliable, it can be applied to the detection of choline in blood.
