摘要 目的筛选链霉菌宿主并异源表达环肽YM-216391,为组合生物合成新化合物等后续研究奠定基础。方法将包含YM-216391合成整个基因簇的黏粒导入不同的链霉菌中,分析不同宿主YM-216391表达量,并探索培养基配方。结果 YM-216391在白色链霉菌(Streptomyces albus),变铅青链霉菌(S.lividans 1326,S.lividans TK24)和金色链霉菌苏州变种(S.aureus var.suzhouensis var.Yen SP-371)宿主中均可实现异源表达,通过优化培养基配方,YM-216391表达量达到(5.664±1.176)mg/L。结论 YM-216391在不同链霉菌宿主中均可表达,其中在S.albus中表达量最高。 Objective To select Streptomyces hosts for heterologous expression of the cyclic peptide YM-216391 of high yield. Methods Cosmids containing the entire gene cluster of YM-216391 synthesis were imported into different Streptomy- ces hosts, respectively. YM-216391 expression in different was analyzed by HPLC and HPLC-MS, and the culture medium formulation was also explored. Results YM-216391 could be expressed in S. albus, S. lividans 1326, S. lividans TK24 and S. aureus var. suzhouensis var. Yen SP-371, and the YM-216391 heterologous expression level reached (5. 664 ± 1. 176) mg/L by optimizing the medium formula. Conclusion YM-216391 could be expressed in different Streptomyces hosts, among which YM-216391 expression level in S. albus is the highest.
