苦马豆素对家兔脑组织抗氧化功能的影响 被引量:9

Effects of swainsonine on antioxydic function of brain tissue in rabbits

摘要 为了探讨苦马豆素对家兔脑组织抗氧化功能的影响,进一步揭示苦马豆素的毒性作用机理。24只家兔随机分为4组,即对照组、试验Ⅰ组、试验Ⅱ组和试验Ⅲ组。将小花棘豆全草粉碎后,按Ⅰ组添加15%(含苦马豆素30mg/kg)、Ⅱ组添加30%(含苦马豆素60mg/kg)、Ⅲ组添加45%(含苦马豆素90mg/kg)的比例制作混合饲料,饲喂至出现中毒典型临床症状为止。攻毒后14,35,70d每组随机采集2只家兔的肝脏组织,检测SOD、GSH-Px、CAT、NOS活性及MDA、NEFA、·OH、LPO、NO和Glu含量的变化。结果显示,与正常对照组相比,试验组家兔肝脏SOD、GSH-Px、CAT等抗氧化物酶的活性极显著下降(P<0.01),而MDA、NEFA、·OH、LPO、NO和Glu含量极显著上升(P<0.01)。结果表明,不同浓度苦马豆素对家兔脑组织抗氧化功能有显著影响,且具有一定的时间效应和剂量效应关系,长期低剂量摄入苦马豆素可引起家兔不同程度的脑组织损伤。 Twenty four rabbits were divided into 4 groups (the control group and experimental group Ⅰ , Ⅱ, Ⅲ ) to investigate the effects of swainsonine on the antioxydic function of brain tis-sue,and reveal the toxicity mechanism of swainsonine. The dried plant of the Oxytropis glabra DC was comminuted,and different amounts of the grass powder(15% ,30%and 45% corresponding to 30,60,90 mg/kg swainsonine) were mixed with the feeds,respectively. The rabbits consumed the forages freely until typical symptoms were observed. Two rabbits were selected randomly from each group on the 14th,35th and 70th day, respectively, the liver was collected and the activity of superoxide dismutase (SOD),glutathione peroxidase (GSH-Px),catalase(CAT) and nitric oxide synthase(NOS) ,the contents of maleic dialdehyde (MDA),renal free fatty acids(NEFA),hydrox-yl radical( · OH),lipofusein(LPO),nitric oxide(NO)and glutamie acid(Glu). The results demon-strated that the activity of SOD, GSH-Px and CAT in experimental groups were significantly lower than those of normal control group(P〈0.01),and the contents of MDA,NEFA, · OH,LPO,NO and Glu in experimental groups were significantly higher than those of normal control group(P〈 0.01). The results showed that swainsonine has remarkable effects on the antioxydic function of liver in rabbits,in a time-effect as well as a dose-effect relationship. It is concluded that receiving a low-dose of swainsonine for a long time causes damages in the liver of rabbits.
