加酸减压内部沸腾法提取氯化两面针碱及其指纹图谱评价 被引量:3

摘要 为降低酸醇提取过程酸对设备的腐蚀及提高生物碱提取效果,提出了加酸减压内部沸腾提取两面针生物碱的方法,并以指纹图谱的方法对其与其他七种提取方法进行评价。结果表明,在酸乙醇解吸剂和乙醇提取剂质量浓度及用量分别为80%(其中盐酸浓度为0.61%)、40%及3、16mL·g-1,提取温度为50℃,系统压力为0.020MPa,提取时间为3min时,得到氯化两面针碱的提取得率及纯度分别为0.132%和0.657%。分析指纹图谱,发现加酸减压内部沸腾法出峰数量及时间和其他提取方法相同,氯化两面针碱的峰面积与总出峰面积之比较大,提取效果显著,并且加酸减压内部沸腾法能有效减轻提取过程酸对设备的腐蚀。 To reduce acid corrosion of equipments,improve extraction of zanthoxylum nitidum alkaloid technology. The method of acid decompressing inner ebullition was used to intensify the extraction of nitidine chloride, comparing acid decompressing inner ebullition with other seven methods by HPLC fingerprint. Experiment results showed that the yield of nitidine chloride reached 0.132% ,the purity of crude extract reached 0.657%, when the powder of zanthoxylum nitidum was firstly immersed into 3mL·g -1 solvent,which was composed of 80wt% ethanol and 0.61wt% HCl and then was extracted by 16mL·g -1 solvent,which was composed of 40wt% ethanol,at 50℃ for 3min,the pressure was 0.020MPa. Comparing with different extraction technology of HPLC fingerprint,acid decompressing inner ebullition was the same as other methods in the number of peak and type of peak,but its nitidine chloride peak area and the percentage of nitidine chloride peak area were both big, extracting safely and efficiently,which obviously reduced acid corrosion of equipments during extraction process.
