摘要
研究外周血干细胞动员与采集方法。方法 :14例不同类型的肿瘤患者 ,分 2个动员方案组。一组给大剂量依托泊苷 (VP 16 )加rhG CSF动员 ,另一组在常规化疗后直接用rhG CSF动员。使用百特CS 30 0 0plus型血细胞分离机采集 ,每例采集 1— 3次。结果 :2种方案动员 ,均可有效地动员外周血干细胞 ,但VP 16组动员效果明显好于常规化疗组 ,且多数患者第一次采集CD+ 34 细胞量即可达到移植的阈值量 (CD+ 34 细胞 2× 10 6 kg以上 )。 14例患者中 9例已完成干细胞移植 ,血象均在移植后 1月恢复正常。结论 :大剂量VP 16 (6 0 0— 80 0mg m2 )加rhG CSF可有效地动员外周血干细胞 ,且VP 16骨髓外毒性轻 。
Purpose:To study the strateges of mobilization and collection of peripheral blood stem cells. Methods:14 patients with solid tumor were divided to two groups, one was administered with high dose of VP 16 plus rhG CSF for mobilization, and the other was with routine chemotherapy followed by rhG CSF. The peripheral stem cells were collected with Baxter Fenwal CS 3000 plus blood cells seperator. Results:The peripheral haemotopoietic stem cells were successfully mobilized in the two groups. The VP 16 group got better result than the routine chemotherapy group. In VP 16 group, most patients got a threshold number of 2×10 6 CD + 34 cells per kg body weight in the first collections.Of 14 patients,9 received the stem cell transplantationand their blood pictures were restored in one month after the stem cells were reinfused. Conclusions:High dose of VP 16 (600—800 mg/m 2) with the addition of rhG CSF was an effective method for peripheral blood stem cell mobilization. VP 16 had little extramedullary toxicity, and the changes of blood pictures were regular.
