摘要
采用苯酚-硫酸法测定贝类中可溶性糖。探讨提取剂用量、水浴温度、澄清剂用量对贝类中可溶性糖提取的影响,通过单因素试验和正交试验确定贝类中可溶性糖提取的最佳条件为提取剂水用量110 mL,温度90℃,澄清剂用量为5mL乙酸锌和5mL亚铁氰化钾。该方法的精密度和准确度良好,平均RSD值为1.57%,平均回收率为97.76%。
The solube sugar in shellfish was detected by phonel-sulfate method.The amount of water,bath temperature and dosage of clarifier were tested to observe their effect on extraction of soluble sugar.Single factor test and orthogonal experiment were designed to optimize the extraction conditions.The results showed that the best extracting conditions were as followed:the amount of water 110mL,water bath temperature 90℃and the dosage of clarifier 5mL zinc acetate with 5mL potassium ferrocyanide.The accuracy and precision of this method is good,with the average RSD 1.57% and the average recovery 97.76%.Then the contents of soluble sugar of several shellfishes were detected in this extraction conditions.
