摘要
目的:测定市售川乌不同品种中总黄酮的含量并比较其差异。方法:利用超声波提取法,在温度55℃、时间40min、以70%乙醇为提取溶剂、物料比为1∶20(m/V)的条件下提取10个样品中的黄酮类化合物;以芦丁为对照品,在510nm波长处,采用紫外分光光度法测定总黄酮的含量。结果:芦丁的质量浓度在0~0.0988mg/ml范围内与其吸光度呈良好的线性关系(r=0.9998);精密度、稳定性、重复性试验的RSD均<2%;平均加样回收率为97.93%,RSD=1.55%(n=6)。10个市售川乌样品中,总黄酮质量分数最高的是8号,其质量分数极显著高于10、4、6、7、2、1、9、3号,显著高于5号;3号川乌的总黄酮质量分数最低,但与7、2、1、9号川乌比较差异无统计学意义。结论:市售不同品种川乌中总黄酮的质量分数存在明显差异,临床应注意甄别使用。
OBJECTIVE: To determine and compare the contents of flavonoids from different types of marketable Aconitum car- michaelii. METHODS: Using ultrasonic extraction, the flavonoids compounds were extracted from 10 samples at 55 ℃ for 40 min with the material ratio of 1 : 20 (m/V)using 70% ethanol as solvent. The content of flavonoids was determined by UV spectropho- tometry at 510 nm, using rutin as substance control. RESULTS: The linear range of rutin were 0-0.098 8 mg/ml (r=0.999 8) with an average recovery of 97.93% (RSD=1.55%, n=6). RSDs of precision, stability and reproducibility tests were all lower than 2%. Among 10 types of marketable A. carmichaelii, the content of flavonoids in No. 8 sample was the highest, which was very sig- nificantly higher than in No. 10, No.4, No. 6, No.7, No.2, No.l, No.9 and No.3 samples,and significantly higher than in No.5 samples; the content flavonoids in No. 3 sample was the lowest, which was not significantly different from No. 7, No.2,No. 1 and No.9 samples. CONCLUSIONS: The contents of flavonoids are different significantly in different types of marketable A. carmichae- lii, which should be used after identified in the clinic.
